Detection of TIMP2 protein in plasma between the cord blood and peripheral blood of the elder

Abstract:

Objective

To explore the effects of different treatments on the detection of TIMP2 protein in umbilical cord blood or peripheral blood of elder. The establishment of a method for detecting the stability of TIMP2 protein in human plasma provided a theoretical basis for the possibility of combining stem cells to anti-aging.

Methods

15 umbilical cord blood samples and 15 peripheral blood samples of elder were collected. The effects of final detection of TIMP2 protein in plasma were investigated, which was used different centrifugal forces, different storage temperatures and different test kits. The difference between the two groups were compared by one-way ANOVA and independent sample t-test.

Results

There was no significant difference in TIMP2 protein content among 1?000×g centrifugal force, 1?500×g centrifugal force and 2?000×g centrifugal force. There was no significant difference in TIMP2 protein content among 4℃, -20℃ and -80℃ storage temperature. The R&D Systems, Abnova and Abcam test kits did not significantly affect the final detection of TIMP2 protein in whole blood. Moreover, the contents of TIMP2 protein in umbilical cord blood (147.00±15.92) pg/ml were significantly higher than that in the peripheral blood of the elder (79.56±10.80, P?< 0.01) .

Conclusion

Different centrifugal forces, different storage temperatures and different test kits did not affect the final detection of TIMP2 in the cord blood and peripheral blood of the elder. Moreover, the contents of TIMP2 protein in the cord blood were significantly higher than that in the peripheral blood of the elder, which indicated that TIMP2 protein had a direct relationship of aging. It is of a potential value in anti-aging combined with the stem cells.
